土壤微生物的分离纯化及理化性质的研究(微生物的综合实验论文)

从土壤中分离和纯化细菌、放线菌

摘要: 土壤是微生物生活的大本营，是寻找有重要应用潜力的微生物的主要菌源。通过采用无菌操作技术，先用稀释法制备土壤的菌悬液，然后结合选择培养基用平板划线法分离出纯菌株，最后用斜面接种的方法对菌株进行保存。通过观察和描述培养特征对菌株所属类群进行初步鉴定，对放线菌进行镜检，对细菌进行革兰氏染色方法观察，以及进行细菌最适合pH生长的初步研究。观察到放线菌的孢子结构；得知该细菌为革兰氏阴性菌，且研究得知该细菌最适合生长的pH是7.48。

Separate and purify the bacteria and

actinomycetes from the soil

Abstract: The soil is the microorganism life supreme headquarters, and is the main fungus source to seek the important application potential microorganism. Through the aseptic technique, uses the dilution method preparation soil the fungus to hang the fluid first, then the union choice culture medium separates the pure strain with the plate streaking law, finally uses the incline vaccination the method to carry on the preservation to the strain. Through the observation and describe training characteristics on the preliminary appraisal belongs strains groups, actinomyces on anti-bma bacteria to gram stain method observation, and the most suitable for the growth of bacteria pH preliminary study. We have observed the actinomycetes’ spore structure and known this bacterium is a kind of the Gram-negative bacteria. Besides, through the research we have known that the most suitable pH for the bacterium to grow is 7.48.
