摘 要：目的 通过通心络胶囊对载脂蛋白E基因敲除〔ApoE（-/-）〕模型组小鼠基质金属蛋白酶-1（MMP-1）、基质金属蛋白酶-9（MMP-9）和基质金属蛋白酶组织抑制剂1（TIMP1）水平等指标的干预变化，探究其治疗冠心病（CHD）的机理。方法 将40只ApoE （-/-）小鼠作为实验组，建立冠状动脉粥样硬化（AS）模型，随机分为模型组等5组，分别给予辛伐他汀及不同剂量的通心络胶囊进行干预，另选8只同系的小鼠作为正常对照组。通过对模型组小鼠血清MMP-1、MMP-9 和TIMP1水平的观察，比较分析通心络胶囊对ApoE （-/-）小鼠冠状AS的调节作用并探讨其机理。结果 通心络胶囊高剂量组小鼠血清MMP-1和MMP-9水平均低于模型组，而TIMP1水平高于模型组，与模型组相比，均有统计学意义（P<0.05或P<0.01）。通心络胶囊中剂量组小鼠血清MMP-9水平低于模型组，与模型组相比有统计学意义（P<0.05）。结论 通心络胶囊具有抑制血管重构和稳定斑块的作用。 关键词：通心络胶囊；ApoE（-/-）小鼠；冠状AS；MMP-1；MMP-9；TIMP1；干预

Abstract Objectives To investigate mechanism of Tongxinluo Capsule managing coronary heart disease (CHD) by observing changes resulting from intervention of apolipoprotein E gene knocking out [ApoE (-/-) ] MMP-1, MMP-9 and TIMP1 in mice model. Method 40 ApoE (-/-) mice, as experimental group, were used to establish coronary arteriosclerosis (AS) model and then were randomly divided into 5 model groups. Simvastatin and Tongxinluo Capsule at different dosage were administrated to mice respectively as intervention and 8 mice of same kind were selected as control group. Tongxinluo Capsule's effect in regulating coronary AS in ApoE (-/-) mice and its mechanism were compared and analysed during the experiment by observing serum MMP-1,MMP-9 and TIMP1 levels in model mice. Results in group administrated high dose Tongxinluo Capsule, serum MMP-1 and MMP-9 level were lower than that in model group without intervention, but TIMP1 was higher. The difference is significant statistically (P<0.05 or P<0.01). In the group administrated moderate dosage Tongxinluo Capsule, the serum MMP-9 was lower than that in model group without intervention, which also has statistical significance (P<0.05). Conclusion Tongxinluo Capsule is of some effect in the inhibition of vascular remodeling and stabilization of plaques.
